一株磺化钻井液降解菌的筛选鉴定及其降解特性

Screening, evaluation and degradation property of the degrading bacteria suitable for sulphonated drilling fluid

  • 摘要: 为了筛选磺化钻井液高效降解菌株,以新疆某磺化钻井液伤害土壤为菌源,磺化钻井液为唯一碳源,通过富集驯化分离筛选出一株降解磺化钻井液效率较高的菌株Xh8,经鉴定为Bacillus flexus。经生理生化及16S rRNA基因序列分析确定了其分类地位,并进行了单因素和正交试验以确定其降解特性。单因素试验表明Xh8生长最佳条件为:pH值为9.5、盐质量分数为4%、转速为150 r/min、温度为35 ℃、接种量为10%、N源蛋白胨、P源K2HPO4。正交试验表明磺化钻井液COD降解最佳条件为:pH值为9.5、盐质量分数为4.5%、转速为150 r/min、温度为35 ℃。培养7 d,磺化钻井液COD降解率为22.9%。试验结果表明,Xh8可作为磺化钻井液降解的优良备选菌株。该研究可为进一步治理受到磺化钻井液伤害的地层提供理论指导。

     

    Abstract: In order to screen out the bacteria strain that can degrade sulphonated drilling fluid efficiently, a series of studies were carried out with the sulphonated drilling fluid as the only carbon source and some soil contaminated by the sulphonated drilling fluid in Xinjiang as the bacteria source. The bacteria strain Xh8 that can degrade sulphonated drilling fluid more efficiently was screened out by means of enrichment, acclimation and separation. And according to the evaluation, it is Bacillus flexus. Then, its classification status was determined by virtue of physiological & biochemical and 16S rRNA gene sequence analysis. In addition, its degradation property was figured out through single-factor experiment and orthogonal experiment. The single-factor experiment results show that the growth of Xh8 is optimal in the condition of pH value 9.5, salt mass fraction 4%, rotation speed 150 r/min, temperature 35 ℃, inoculation amount 10%, N-source peptone, and P-source K2HPO4. The orthogonal experiment results show that the optimal COD degradation condition of sulphonated drilling fluid is pH value 9.5, salt mass fraction 4.5%, rotation speed 150 r/min and temperature 35 ℃. After being cultured for 7 days, the COD degradation rate of sulphonated drilling fluid is 22.9%. The experimental results indicate that Xh8 can be taken as the preferred bacteria strain for the degradation of sulphonated drilling fluid. The research results can provide the theoretical guidance for the further treatment of the strata contaminated by sulphonated drilling fluid.
